We first see Jo when the boys visit her mom's, Ellen's, Roadhouse.   Throughout the first part of Season 2 we get to know a lot about how badly Jo wants to become a hunter and even get a chance to see her in action in the episode "No Exit" where she goes up against the ghost of HH Holmes along side Dean and Sam.   This is also the episode where she disappears after hearing that John Winchester caused the death of her father.  She does not reappear until the episode "Born under a bad sign" When Sam pays her a visit at her new bar.  Here we also learn that Jo is harboring feelings for Dean Winchester along with that fact that John Winchester did not have a choice in the actions he took with her father.
