Before Mary married John she made a deal with Azazel (Yellow-Eyed Demon) that on the sixth month of her youngest child's life he would be allowed access to the nursery with no interruptions.  We do not know how the deal originally went down but we do know that this deal was made to bring John Winchester back to life at the end of the Episode "In the Beginning".

Mary died the night of Sam's sixth month "birthday" because she interrupted Azazel while he was visiting Sam.  This action set the course that Sam and Dean now are traveling.
